ニューヨークのコロンビア大学で超常現象や幽霊・霊体の研究を行っていたピーター・ヴェンクマン博士ら冴えない研究者3人。ある日、一方的に研究費を打ち切られたことをきっかけに借金に借金を重ね、科学的に超常現象全般を扱い幽霊退治を行う会社「ゴーストバスターズ」を開業。当初は資金もなく依頼もゼロに近かったが、とあるホテルでの幽霊退治をきっかけにビジネスは大当たり、メディアや行政からも注目視される。

破壊神からニューヨークを救った彼らだったが、破壊した建物などの賠償金を市と州より請求された上、数々の違法行為の責任を押し付けられ「ゴーストバスターズ」社は破産してしまい、依頼もぱったりと来なくなった。市民も彼らと超常現象を忘れ去り、4人組もそれぞれ別の道を歩んでいた。

1971年、若い夫婦と5人の娘のペロン一家は、ロードアイランド州ハリスヴィルの田舎にある、いささか古いが屋敷のように広い部屋を持つ一軒家を購入した。念願のマイホーム購入を喜ぶペロン一家であったが、入居した翌日から奇怪な現象が次々と発生するようになる。 ついに娘たちにもその危害が及ぶに至って、ペロン夫妻の妻キャロリンは、超常現象研究家夫妻として名高いウォーレン夫妻に助けを求める。夫のエドはカトリック教会が唯一公認した非聖職者の悪魔研究家であり、妻のロレインは透視能力を持っている。 ウォーレン夫妻の調査の結果、その一軒家には戦慄すべき血塗られた過去がある事が判明した。ウォーレン夫妻はペロン一家を救うべく尽力するが、霊力の強さと邪悪さはウォーレン夫妻の想像をはるかに超えるものであった。

若手映像作家のアレックスは、ヤラセ番組として葬り去られたかつての人気テレビ番組「グレイヴ・エンカウンターズ」が、実は本物だったのではないかとの疑いを抱いていた。そして独自の調査でその思いを強くした彼は、「グレイヴ・エンカウンターズ」の真実を解明するドキュメンタリーの製作を思いつく。

A few months before its demolition, four young men came to investigate the clinic, determined to discover the truth about rumors of strange and paranormal events, recording everything with which they encounter.
