ロバート・ネビルは優れた科学者だが、彼でさえ、その恐ろしいウイルスを食い止めるすべを知らなかった。それは誰にも封じることができず、治すこともかなわず、しかも、人間が作り出したウイルスだった。なぜか免疫があったネビルは、荒れ果てたニューヨークで――あるいは世界で――ただ１人生き残っている。この３年間、彼は毎日几帳面に無線でメッセージを流し続け、必死にほかの生存者を捜してきた。だが、確かに彼は"１人"ではなかった。ウイルスに侵され、変異した人間たちが闇に潜み、彼を見つめていたのだ。ネビルを駆りたてているのは唯一残った使命、それは、彼自身の免疫のある血液を使ってウイルスの影響を改善する方法を見つけること。しかし、彼には圧倒的多数の敵が迫り、時間は刻々となくなっていく...。

A deadly virus has spread across the globe. Contagion is everywhere, no one is safe, and no one can be trusted. Four friends race through the back roads of the American West on their way to a secluded utopian beach in the Gulf of Mexico where they could peacefully wait out the pandemic. Their plans take a grim turn when their car breaks down on an isolated road starting a chain of events that will seal their fates.

26年前に爆発事故を起こし問題となったロシアのチェルノブイリ原子力発電所。今では周辺の街も含め廃墟とかした土地になっている。物語は、今なお放射能の残る踏み込んではいけないチェルノブイリの居住地を訪れた観光客たちが、子供のような影を見かける。興味本意で調べはじめるが、一人また一人と得体の知れない影に襲われていく。

太陽フレアの影響で地上の文明が崩壊し、地下シェルターに逃れて生き延びたロボット技術者のフィンチは、愛犬と自ら作り出したロボットとともに、安心して暮らせる新たな家を見つけるための旅に出る。さまざまな危険や困難が待ち受けるその旅を通して、人間と犬とロボットという個性的な組み合わせの一行は、次第に家族のように心を通わせていく。

症状がレベル5まで進行した感染者は、凶暴化して人間を襲い始めるという危険なウイルスが蔓延するアメリカ。ある秘密と目的を抱えながら、CDC（疾病管理予防センター）のドクターとして、感染者から未感染者を守る救助チームへの配属を希望した女性ローレン。彼女の狙いは、本来感染者は救助不能とされるところ、ドクターなど貴重な人材の場合に限り、レベル1までなら治療できる特効薬を使えるという特権にあった。
