ダメな新入生を立派な警官に育てることに一所懸命なラサール校長のもとに、弟のラサール署長から電話がかかった。弟のラサールは、全米で最悪の犯罪地区の署長をしている。彼のため、そして、その地区の凶悪な暴走族を一網打尽にするために、校長は、ポリスアカデミーの卒業生の中から特に優秀な警官を選抜する。そこで選ばれたのが、マホニー、怪力超人ハイタワー、拳銃狂タックルベリー、物まね狂、通称人間効果音ラーヴェル、内気なフックス、脱ダメ亭主のダグラスの6人だった。

仲間の裏切りで逮捕される直前、ダイヤモンド“ブルー・ストリーク”を建設中のビルに隠した宝石泥棒マイルズ。やがて刑務所から出所したマイルズはダイヤを回収しに向かうが、そこは何とロス市警になっていた。宝石を手に入れるため、マイルズは刑事を装って警察署に潜入するが、彼の経験と知識は次々と事件を解決していってしまう。そしてついには麻薬組織を相手にした大捕物に借り出されることに…。

定年を間近に控え、失意の日々を過ごしているラサール校長の姿に見かねたハイタワーたちポリス・アカデミーの卒業生は、永年勤続の表彰をうける校長に同行し、マイアミへ向かうことになった。一行は、ラサール校長の甥である刑事部長ニックに出迎えられるが、空港で校長はトニーを主犯とするダイヤモンド強奪一味と自分のバッグを取り間違えてしまう。宝石を取り戻すため一味は校長を誘拐するが、本人はそれを警察の捜査訓練の一環と思い込み、おまけに校長の後釜をねらってマイアミにやってきたハリス署長がそれにからんできて、事件はてんやわんやの大騒動となる。
