天才物理学者として将来を期待されるスティーヴン・ホーキングはケンブリッジ大学大学院に在籍中、詩について勉強していたジェーンと出会い恋に落ちる。その直後、彼はALS（筋萎縮性側索硬化症）を発症し余命は2年だと言われてしまう。それでもスティーヴンと共に困難を乗り越え、彼を支えることを選んだジェーンは、二人で力を合わせて難病に立ち向かっていく。

他人とのコミュニケーションが苦手な半面、数学の理解力に関してはずば抜けた才能を持つ少年のネイサン。そんな彼の唯一の理解者であった父親が、ある日、彼の目の前で事故死を遂げ、ネイサンはますます周囲に心を閉ざすようになってしまう。母親のジュリーは、数学教師のマーティンにネイサンの個人指導を依頼。そのかいあってネイサンは国際数学オリンピックの英国代表チームの一員に選抜され、仲間たちと強化合宿に参加する。
