冴えないバツイチ男ウィリアム・タッカーは、ロンドン西部の町ノッティングヒルで本屋を営んでいる。そんな彼はある日、ハリウッドのスター女優アナ・スコットが自分の店を訪れたことに驚くが、その直後、偶然にも彼女と街角でぶつかった際にオレンジジュースをこぼしてしまい、着替え場所を提供するため自宅に招くことに。彼女の去り際、初対面にも関わらず交わした“シュールだがステキなキス”から、ノッティングヒルの本屋とビバリーヒルズのスターの恋が始まった…

幼くして両親を亡くし、伯父夫婦のもとで大切に育てられたピーター。高校3年生となった彼は、6歳の頃からずっと思いつづけている隣家のメリー・ジェーンに未だに打ち明けることができないちょっと冴えない高校生。ある日ピーターは親友のハリーから彼の父親ノーマン・オズボーンを紹介される。ノーマンは巨大軍需企業オズコープ社の経営者にして天才科学者。ノーマンはピーターの科学の才能を高く評価、彼に目を掛けるようになる。そんなピーターは、大学の研究所を見学した際、遺伝子組み換えでスーパースパイダーとなったクモに刺されてしまう。その瞬間、ピーターの身体に異変が起こり始める。
