前作で100万もの兵を率いてギリシャ侵攻を図るペルシャ帝国を相手に、300人の精鋭と共に戦いを繰り広げた果てに命を落としたスパルタのレオニダス王。今回の物語は灼熱の門で全滅したレオニダス達と同じ時間の別の戦い、エーゲ海での海戦を軸にしている。 アテナイのテミストクレス将軍は、パン屋、陶工、詩人といった一般市民から成るギリシャ連合軍を率いてペルシャ帝国海軍に立ち向かっていく。 ペルシャ帝国の海軍指揮官アルテミシアらと拮抗する中、ついに大海原を舞台にした最終決戦を迎えることに。

1873年、アリゾナ準州、一人の男（ダニエル・クレイグ）が真昼の砂漠で目ざめた。男は記憶を失なっており、右腹に深い傷を負っていた。身につけているのはボロボロの下着と、左手に嵌った奇妙な金属製の腕輪のみ。近くには見知らぬ女性が写った小さな写真が落ちていた。男は死に物狂いで腕輪を外そうとするが、どうしても外れない。そこに3人のならず者達が通りかかり、男が丸腰であるのを見て、追いはぎを行なおうとするが、逆に男は瞬く間に3人のならず者達を殺傷し、彼らの衣服、武器、荷物そして馬を奪う。
