In 1999, Star Wars fanatics take a cross-country trip to George Lucas' Skywalker Ranch so their dying friend can see a screening of The Phantom Menace before its release.

フィラデルフィアの一流法律会社に務めるアンドリュー・ベケット（トム・ハンクス）は、ある日突然エイズと宣告され、ウィラー社長（ジェイソン・ロバーズ）に解雇される。不当な差別に怒ったベケットは、損害賠償と地位の保全を求めて訴訟を決意。だが、次々と弁護を断わられた彼は、以前敵同士として渡り合ったやり手の弁護士ジョー・ミラー（デンゼル・ワシントン）を訪ねる。ミラーはエイズに対して、抜きがたい恐怖を感じていた。しかし、世間の冷たい視線に対しても毅然と対処し、熱心に資料を漁るべケットの姿に、ミラーの心は動かされる。ミラーは弁護を引き受け、母のサラ（ジョアン・ウッドワード）をはじめ、ベケットの肉親たちは彼に熱い支援を約束する。解雇から7カ月後、〈自由と兄弟愛の街〉フィラデルフィアで注目の裁判が開廷した。ミラーは解雇が明らかな法律違反だと主張したが、対する会社側の主任弁護士ベリンダ（メアリー・スティーンバージェン）は、彼の弁護士としての不適格性を激しく突く。予断を許さぬ裁断の行方と並行して、ベケットの症状は次第に悪化していく。裁判を優先させて本格的治療を先に延ばそうとする彼に、恋人でライフパートナーのミゲール（アントニオ・バンデラス）は苛立つ。ベケットは恋人のため、自分のためにパーティを開く。遂にベケットは裁判中に倒れ、病院に運ばれた。ミラーは原告側の勝訴の報を、ベッドの上のベケットに告げる。数日後、大勢の人々に見守られながらベケットは静かに息を引き取り、ミラーはかけがいのない友の死を実感した。
