The world now knows the story of this small town massacre, seven Community Service workers and many others, on one horrendous weekend lead by officer Bob Butterfield, an old childhood tormentor of psychiatric serial killer Billy Fouls, but they don't know that you can't kill Billy. Getting ready to take a break from reality for a while, Officer Bob Butterfield agrees to one last job, a few hundred miles away, at an abandon amusement park on a private island. With two x Marines joining on the journey as Community Service Officers, no better way to keep the ten Community Service Workers in line, right? The owner of the amusement park, a cracked out clown, and his pal, "a little person", are always on premise and loves to put on a good show for the very few who find this island. The question is, will they be the ones putting on the show.

CIAのヴァレリー・プレイム（ナオミ・ワッツ）と元ニジェール大使で夫のジョー・ウィルソン（ショーン・ペン）は、イラクに核開発計画がないことを政府に報告する。しかし2003年、ブッシュ政権がイラクに宣戦布告。ジョーがニューヨークタイムズ紙にイラクにおける調査報告を寄稿したことから、報復としてヴァレリーが諜報員であることがアメリカ中に公表されてしまい……。
ボンドは、石油王のロバート・キング卿に返金される大金を奪取する事に成功。キング卿が大金を確認するためMI6に訪れる。ボンドがMへの報告中、その大金に爆弾が仕掛けられていることに気づき止めようとするが間に合わず、キング卿は爆死した。ボンドは犯人と思われる女暗殺者を追うが、激しいボートチェイスの末、女暗殺者は気球と共に自爆する。 真犯人は009に銃弾を頭に撃ち込まれるも死なず、そのせいで痛みを感じない体となった不死身のテロリスト「レナード」だと睨んだMI6は、キング卿の娘であり、レナードに誘拐された経験があるエレクトラ・キングが次に狙われると判断。ボンドに彼女の警護を命じる。 そんな時、レナードによって核弾頭が盗まれ、キング社の石油パイプラインにその核弾頭が仕掛けられる。
ブレイドはヴァンパイアと人間との間に生まれた混血で、人間を脅かすヴァンパイアを抹殺するために闘うヴァンパイア・ハンターだ。彼は母親を死に追いやった宿敵のヴァンパイア、フロストを追っていた。フロストは世界征服を狙い、暗黒院の書庫で古代予言書「マルガの書」をコンピュータで解読し、12人のヴァンパイアを生け贄に全能の力を得ようと企んでいた。ブレイドは彼らのアジト暗黒院に潜入、フロストがマルガを呼び起こす儀式を阻止しようとする。
社会に紛れたヴァンパイアと戦う闇のヒーロー、ブレイドは敵のひとりを射殺するが、相手がヴァンパイアの奴隷と化した人間だったことから殺人犯として拘束される。相棒ウィスラーの娘アビゲイルと武器の達人ハンニバル・キングに救われたブレイドは、2人の案内で人間のヴァンパイア・ハンター組織“ナイトウォーカー”と合流する。組織は彼に、かつてはドラキュラと呼ばれたヴァンパイアの始祖・ドレイクが復活したことを告げる。
ボンドは北朝鮮側の非武装地帯にある基地で、アフリカから不正輸出されたダイヤモンドと引き換えに武器の密輸を行なっていたムーン大佐抹殺の任務を遂行した直後、彼の父であるムーン将軍に捕らわれ、長きに渡る監禁・拷問を受ける。 14ヶ月後、中華人民共和国の諜報員3名を殺害したのち、逮捕されたムーン大佐の側近のザオとの捕虜交換が行われ、ようやくMのもとに戻れたボンドは思いがけない言葉を耳にする。それは00（ダブルオー）ナンバーの剥奪だった。1週間前に北朝鮮内部に潜り込んでいたアメリカの工作員が処刑され、ボンドが居た収容所から情報が発信された事から、北朝鮮での拷問でボンドが機密事項を洩らしたのが原因だと疑うアメリカはこれ以上の情報漏れを恐れザオとの交換でボンドを連れ戻したのだという。ボンドを疑うMは、ボンドの00ナンバー剥奪という決断に至ったのだ。 この結果に納得のいかないボンドは、自らのプライドと00ナンバーを取り戻すべく単身でMたちのもとから脱出。香港を拠点に活動する中国の諜報員ミスター・チャンの協力のもと、ザオがキューバに潜伏中との情報をつかみ、キューバに飛ぶ。そこでアメリカ国家安全保障局（NSA）の諜報員ジンクスと遭遇する。
世界は冷戦真っ只中。ジェームズ・ボンドの007復帰トレーニングから物語は始まる。マティーニとフォアグラ、キャビアで怠惰になった身体を鍛えなおすことを新着したMに命令された007。たまたま本部から斡旋されたロンドン郊外の治療施設でボンドはスペクターの女殺し屋ファティマ、そしてアメリカ空軍に所属するジャックの秘密特訓を目撃する。そしてその数日後、ジャックは米空軍より核弾頭搭載巡航ミサイルを2機盗むことに成功する。 この事件を追うボンドは地中海へ向かい、世界的な大富豪のラルゴをマークする。世界の海を豪華クルーザー「空飛ぶ円盤号」（クルーザー全長87m、アドナン・カショーギ所有）で移動しながら慈善活動を行なうビジネスマンだが、裏の顔は秘密結社スペクターのNo.1（スペクターNo.2＝首領は白猫を抱くブロフェルド）であった。そして彼のそばにはジャックの妹であるドミノがいた。

内部汚職の根を絶ち、殺しのライセンス・00（ダブルオー）を得たボンドはマダガスカルである男の監視をしていた。その男の携帯電話から得た情報、「エリプシス」の糸を手繰りバハマへ。そこでメッセージの送信者で、武器の売人のディミトリオスおよびその妻ソランジュと接触。マイアミ国際空港で披露される超大型旅客機の爆破計画を知る。この計画に絡んでいるとしてル・シッフルという男の情報をMから得る。ル・シッフルは世界各国のテロ組織から預かった資金をマネーロンダリングしつつ運用しており、旅客機製造会社の株のカラ売りを仕込んだ上で、同社が製造した超大型旅客機をお披露目式で爆破して、巨額の利益を得ようとしていた。しかし爆破をボンドに阻止されたことで大金を失い、テロ組織に返却する資金を取り戻そうとモンテネグロの「カジノ・ロワイヤル」にて開催されるポーカーゲームに参加することになった。ボンドはそのゲームに参加し、国の資金を使ってテロ資金稼ぎを阻止するよう命じられ、監視役として送られて来た金融活動部（FATF）のヴェスパー・リンドとともにゲームへ挑む。
メカの天才スカッドを新たな相棒にしたブレイドは、ヴァンパイアのアジトを襲撃。とらわれた相棒ウィスラーを取り戻すがその矢先、今度は逆にブレイドの隠れ家が発見されてしまう。戦闘態勢を敷くブレイドたちだが、現われたヴァンパイア部隊は休戦を申し入れてきた。人間もヴァンパイアも根絶させたい新種族リーパーズに対抗するため、一時的に手を結ぼうという。ブレイドはウィスラーら仲間に反対されるが、提案を受け入れる。
