徳川八代将軍・吉宗の時代。ある日、大奥お年寄・藤尾宛へ、紋之助という役者の入ったつづらが届けられた。早速大目付・矢野伝八郎の取調べが始まり、藤尾には身の覚えがないことであるが、御用屋敷にお預け、藤尾に仕えるおこうは江戸城追放の処分となった。そして藤尾の後任には竹川が抜擢され、竹川は自分の下で働くお静に吉宗の伽を勤めさすようにした。一方、藤尾の下にいたおるいとおせちは、この度の一件で誰が藤尾を陥し入れようと計ったのか突きとめようと苦心していた。数日後、おこうが大奥の入口を警護する成島伊助から紋之助の相手をしていたのは「左の内股に大きなほくろのある女中」ということを聞き出した。その情報はおるいとおせへ早速知らせた。ある日、諸国大名より献上の美女くらべが行われ、その中で吉宗は金髪娘サリーを大そう気に入り、毎晩のように可愛いがった。ところがサリーの世話役をしていたおちせが吉宗の眼にとまってしまった。おちせには信次郎という婚約者がいたが、吉宗は許さず、半ば強引におちせを伽へと招いた。ところが吉宗がおちせの股にあるホクロを見付けると、お静にも同じところにホクロがあると言った。おちせには真相の全てが解った。藤尾の名を限っ騙ったのはお静で、竹川と矢野が仕組んだことだったのである。尚もおちせが驚いたことにサリーは老中から藤尾の一件の調査の為に大奥に送りこまれたのであった。やがて藤尾は大奥へ戻り、おちせと新次郎は目出たく所帯をもつことになったのである。
