1932年、品格が漂うグランド・ブダペスト・ホテルを仕切る名コンシェルジュのグスタヴ・Hは、究極のおもてなしを信条に大勢の顧客たちをもてなしていた。しかし、常連客のマダムDが殺されたことでばく大な遺産争いに巻き込まれてしまう。グスタヴは信頼するベルボーイのゼロと一緒にホテルの威信を維持すべく、ヨーロッパ中を駆け巡る。
