あるブリーフケースを盗むよう謎の女性から指令を受け、東京発京都行の高速列車に乗り込んだ殺し屋・レディバグ(ブラッド・ピット)。ブリーフケースを奪って降りるだけの簡単な任務のはずだったが、疾走する車内で次々に殺し屋たちと遭遇してしまう。襲い掛かってくる彼らと訳も分からぬまま死闘を繰り広げる中、次第に殺し屋たちとの過去の因縁が浮かび上がってくる。
