ニューヨークで雑誌の調査員として働くソフィは、婚約者のヴィクターとイタリアのヴェローナに婚前旅行でやって来る。ところが、レストランの開店を予定しているヴィクターはソフィそっちのけで食材探しに夢中。仕方なく、一人で“ジュリエットの家”を訪れたソフィ。偶然にも、壁の中にあった一通の“ジュリエット・レター”を見つける。それは、50年前にイタリアを訪れ、そこで出会った青年ロレンツォと恋に落ちた英国人女性クレアが書いたものだった。その手紙にソフィが返事を書いたところ、それを受け取ったクレアが孫のチャーリーを伴ってはるばるイタリアまでやって来た。ソフィはクレアの話を聞くと、ロレンツォを捜し出そうと提案、こうして３人で50年前の初恋の相手を捜す旅が始まるのだが…。
