いろいろなガジェットを使い犯罪を繰り返すバルタザール・ブラットを逃したことで、反悪党同盟を追い出されてしまうグルー。意識消沈する中、生き別れになっていた双子の兄弟であるドルーの存在が判明する。対面を果たしたグルーは、豊かな金髪に輝く笑顔で父親からばく大な遺産を受け継いだ、自分とは違い過ぎるドルーに驚く。一方、グルーが悪の道に戻らないことがわかり彼と決別したミニオンたちは、新たなボスを探す中で思わぬ事態になる。
