ルーブル美術館の館長が殺され、宗教象徴学の権威ラングドン教授に捜査協力が依頼される。遺体はダ・ヴィンチの素描「ウィトルウィウス的人体図」を模した姿勢を取らされ、周囲には暗号が記されていた。担当のファーシュ警部は、暗号が“ラングドンを捜せ”と示していたことから教授を疑うが、館長の孫ソフィーは教授の無実を信じ、彼を現場から逃がす。2人は警察に追われながら、暗号の真意と真犯人を捜し始める。
