Two proud and estranged brothers, a disputed inheritance and a past that comes back. ハーバード大学の宗教象徴学者ラングドン教授は、数日分の記憶を失った状態で、フィレンツェの病院で目を覚ます。謎の襲撃者に狙われたラングドンは、美しい女医シエナ・ブルックスに助けられて病院を脱出。何者かから追われる身となったラングドンとシエナは、生物学者ゾブリストが人類増加問題の解決策として恐ろしい伝染病を世界に広めようとしていることを知る。そしてゾブリストが詩人ダンテの叙事詩「神曲」の「地獄篇」になぞらえて計画を実行していることに気づき、阻止するべく奔走する。
