真っ赤でピカピカなボディがトレードマークのレース・カー“ライトニング・マックィーン”は、レースでチャンピオンを目指す人気者。ある日ルート66号線沿いにある田舎町“ラジエイター・スプリングス”に立ち寄り、住民たちと触れ合ったことで、自分の人生には何かが失われていたことに気が付く。

モンスターズ株式会社は、住民全員がモンスターのモンスター・シティと呼ばれる町にあった。サリーと相棒のマイクら社員は毎晩町のエネルギー源である子どもたちの悲鳴を収集するために人間界へと出向いていたが、ある日、幼い女の子ブーが迷い込んで来る。何より人間の子どもに恐怖を抱いていたモンスターたちは、パニックに陥り……。

ある日、ウッディがオモチャ屋のアルにさらわれてしまう。ビンテージ品としての価値に目を付けたアルは、彼を博物館へ売りつけようと奔走。ウッディの友人であるバズたちは、彼を助けるためアルの行方を追う。

池の中に浮かぶ島、アント・アイランドには主にアリたちが暮らしていた。夏になると島にバッタたちがやってきて、秋までの間にアリたちに食料を集めさせる仕事をさせていた。まるで農民たちに年貢を納めさせるかのように。奇抜な発明家・フリックはその発明品があまりに危険なことからアリたちに「歩く災難」とまで言われていた。彼は刈り取り機を発明したが、失敗してしまい集めた食料が川に流れてしまった。フリックは責任をとるためにバッタたちと対決することを決意し、手助けしてくれる強い用心棒を探すために都会に出ることをアリ達と約束したのだった。都会へ向かう道中、あるサーカス団が公演を行っていた。団員たちは公演中に起きたある事件がきっかけで全員解雇され、職を失ったのだった。
