18년만에 출감한 엘우드 브루스(Elwood Blues: 댄 애크로이드 분). 검은 양복에 중절모, 그리고 선글라스. 그의 모습은 그대로이지만 엘우드는 모든 것이 변했음을 깨닫는다. 고아원 시절부터 고락을 같이했던 제이크는 감옥에서 숨진지 오래고 그의 밴드는 해체됐다. 시카고의 고아원 출신인 엘우드는 어릴 때부터 자신을 돌봐준 메어리 수녀가 운영하고 있는 병원을 찾아간다. 그곳에서 엘우드는 말썽장이 고아소년 버스터(Buster: J. 에반 보니팬트 분)를 만나고 버스터는 엘우드를 따라가고 싶어한다. 경찰 서장으로 근무하는 옛친구의 동생 케이블(Cabel Chamberlain: 죠 모튼 분)을 찾아갔다가 우연히 그의 지갑을 손에 넣게 된 엘우드는 자신이 점찍어둔 자동차를 사서 멤버를 구하기 위한 길에 오른다. 하지만 새로운 멤버를 결성하기 위해 여러 사람들을 차례로 찾아가지만 일은 쉽지 않다. 우여곡절 끝에 윌리스 스트립 클럽의 열정적인 비텐더 맥(Mighty Mack McTeer: 존 굿맨 분)을 새로운 멤버로 합류시킨 엘우드는 버스터, 맥과 함께 밴드 경연대회에 출전하기로 한다. 한편, 버스터가 없어진 사실을 안 메어리 수녀는 유괴 혐의로 엘우드를 신고한데다. 지갑이 털렸다고 생각한 케이블은 엘우드를 추적하기 시작한다. 경찰의 추적이 계속되는 가운데 러시아 마피아들과의 한판 대결, 나찌를 신봉하는 KKK단의 훈련과 모임을 엉망으로 만들어 놓는 등 그들은 가는 곳마다 많은 화제와 함께 말썽과 사고를 일으킨다. 결국 대로변 천막에서 열린 부흥회에까지 참가한 이들은 뒤쫓아온 케이블과 경찰들에게 체포되기 직전에 이른다. 그런데 부흥회 중인 천막교회에서 케이블은 믿을 수 없는 하늘의 계시로 놀라운 노래와 춤 실력을 보여주면서 이들의 새로운 멤버로 합류하게 된다. 드디어 엘우드와 맥, 버스터, 케이블로 구성된 환상적인 4인조 블루스 밴드가 탄생한다. 그리고 그들은 밴드 경연대회가 열리는 곳으로 향하는데.
시카고 부근의 한 천주교 고아원에서 자란 형제 제이크(Joliet Jake: 존 벨루시 분)와 엘우드(Elwood: 댄 애크로이드 분)는 검은 양복, 검은 모자, 검은 안경이 트레이드마크인 괴짜들이다. 본업은 리듬앤블루스 밴드 리더였던듯 한데, 오늘은 저간의 곡절로 옥살이를 한 형제 제이크가 모범수로 가석방되는 날이다. 경찰서 폐기처분장비 공매장에서 헐값에 산 중고 경찰차를 몰고 마중 나온 동생 엘우드는 출감한 제이크를 태우고 어렸을때 생활하던 고아원으로 간다. 5천달러의 세금을 며칠 안에 납부하지 않으면 고아원이 교육청에 팔리게 된다는 말을 원장 수녀로부터 들은 블루스 형제는 밴드를 다시 조직하여 큰 공연을 함으로서 5천달러를 마련하겠다는 계획을 세운다. 제이크가 감옥에 들어갈때 각자 살 길을 찾아나서서 지금은 나름대로 자리를 잡고 사는 옛 밴드 멤버들을 우역곡절 끝에 재규합하는데 성공한 블루스 형제는, 오라는 곳 없는 불청객으로 길을 가다 어느 컨트리 앤웨스턴 주점에 들어가 무조건 노래를 부른다. 주점 주인이 연주료 200달러를 주며 밴드가 마신 맥주값 300달러를 청구하자 돈이 없는 블루스 형제는 잽싸게 차에 올라 도주하여 극장 예약업자 모리(Maury Sline: 스티븐 로렌스 분)를 찾아간다. 모리의 약점을 이용하여 5천석 규모의 대극장 공연을 따낸 블루스 형제는 옛 고아원 교사 커티스(Curtis: 캡 콜로웨이 분)와 고아원 아이들의 도움을 받아 하루만에 대규모 선전을 성공리에 마친다. 극장에 구름같이 모여든 청중들은 블루스브라더스 밴드의 노래에 열광한다. 레코드사 사장이 취입 제안을 하면서 선약금 만달러를 내놓기까지 한다.
어릴적 성적 학대를 받은 경험이 있는 레이는 애인 로니가 군에 입대하면서 자신도 절제할 수 없는 성도착증으로 고생한다. 라자루스는 아내가 떠난 이후, 외부와의 소통을 두려워 하며 자신만의 세계에서 하루하루를 보낸다. 어느날 환락의 파티를 마치고 집으로 향하던 레이는 로니의 친구 '길'에게 폭행을 당하고 라자루스 집 앞에 버려진다. 라자루스는 우연히 그녀를 발견하고 집으로 데려와 보살핀다. 레이가 마을에서 소문난 성도착증 환자라는 사실을 알게 된 라자루스는 그녀를 구원하겠다고 결심하고 탈출하려는 그녀를 긴 쇠사슬로 묶어버린다. 처음 라자루스를 원망하고 미워하던 레이도 그렇게 시간이 지나면서 서로에게 마음을 열기 시작한다. 레이의 가슴 속 증오는 조금씩 아물어 가기 시작하고, 라자루스 역시 그녀를 돌보며 자신의 사랑한 음악 연주를 시작한다. 그리고 정신적인 장애로 총을 쏘지 못한다는 이유로 군에서 쫓겨난 레이의 애인 로니가 돌아오고 라자루스에게 총을 겨누게 되는데...
