Uyir Mozhi is a sci-fi film that talks about the miraculous properties of DNA. The story is about 5 boys who fall in love with the same girl and the role DNA plays in their love lives.

The life of Panja, an orphan and a sculptor, revolves around Sathish, Othavada, Kuppan and Sammy, all of whom reside in Mahabalipuram and are close to each other. The only other person Panja considers himself close is Durai, a politician and criminal, who saved him from death when he was a boy. Panja shows his gratitude to Durai by occasionally helping him with his anti-social activities. When Sathish chooses to marry the love of his life, Sangeetha , much against the wishes of their parents, his four friends help him do so. Life is pleasant for a while with Panja too falling in love with Mahalakshmi a college student who moves into the area. However, their lives take a tumultuous turn when the friends chance upon a sleaze video which shocks them out of their wits and results in the death of the newly wed couple. Randy (Harshavardhan Rane) is a guitarist and a noble guy. He is in music field because he loves it, but not to earn money. Sarayu (Vithika Sheru) works for a charity organization and she is in awe with music and inner personality of Randy. Royal Raju (Sree Vishnu) is a street-smart guy from a village who comes to city to enjoy. He joins as an assistant director for a film and he gets attracted to a fashion designer Sameera (Ritu Varma). RJ Arjun works for a FM Radio station and he is a complete flirt. He gets attracted to a Tamil girl Santhi (Sree Mukhi) and wants to bed her. This movie is about how these unrelated love stories culminate simultaneously to tell why men follow their heart and women follow their mind.

The entire story unfolds in a remote village near Vizag called Bheemli. What begins as a soft romance between a poor lad Suri (Nani) and a young girl (Saranya Mohan) who comes to the village soon takes a turn. Suri, along with his childhood friends, yearns to win a Kabaddi tournament. They're known for never managing to win a tournament. They manage to find themselves by chance in a State-level tournament, expected to be knocked out in the very first round. But the bunch of young men who share different thoughts and ideas, with the help of a professional kabbadi coach, emerge as winners. As they proceed, many difficulties are encountered such as Suri having a shoulder dislocation during a match. However, they win all their remaining games and progress to the final. However, during this time, gangsters are searching for Suri.

Balu loves music very much. He wants to become a famous musician. He left his village and came to Hyderabad to fulfill his dream. He rented a house across the house of Venkata Chalapathi's (Mohan Babu) house. Venkata doesn't approve the way of living of the new generation. So when his friend's daughter Sravya and Balu fell for each other, he became complete against of their love. Now the lovers have to overcome the obstacles of Venkata for the sake of their love.
