타이틀롤을 맡은 주인공 EO(당나귀 울음소리에서 착안한 이름)는 동물보호단체에 의해 서커스단으로부터 ‘구조’된다. 이어서 우리는 EO가 농장에서 일하고, 훌리건들에 의해 축구팀 마스코트로 끌려 다니고, 소시지 공장에서 탈출하고, 머나먼 이탈리아의 저택까지 이르는 여정을 따라가게 된다. 동물의 눈으로 바라본 세상은 참으로 낯설고 위협적이며, EO가 거치는 고난의 행군은 예수의 수난에 비견될 만하다.
