파리에서 로마로 가는 급행열차에는 여러 부류의 사람들이 타고 있다. 불륜 관계인 조지와 막스 부인, 영화 배우인 아스터와 누군가를 찾고 있는 저타와 토니, 부유하지만 인색하고 자신의 비서를 학대하는 백만장자 맥베인, 정체를 알 수 없지만 무언가 불안해 보이는 풀 등이 그들이다. 막스 부인과 밀회 여행을 즐기던 조지는 식당차 안에서 아는 사람을 만나게 되자 우연히 옆에 앉아 있던 풀을 자신의 동료인 양 소개하고 객실까지 데려오게 되지만, 다시 막스 부인을 데려오기 위해 풀에게 다른 객실로 가 줄 것을 부탁한다. 하지만 풀은 자신을 쫓는 사람들이 있는 것을 알고 나가지 않으려고 한다. 결국 풀은 조지를 때려 기절시키고, 풀을 쫓던 저타가 객실에 들어와 그를 살해한다. 한편 우연히 풀의 가방과 백만장자 맥베인의 비서의 가방이 바뀌는데 이 가방을 찾기 위해 저타가 다시 이들의 객실에 침입했다가 풀을 쫓기 위해 역시 같은 기차에 탔던 형사 졸리에게 잡히게 된다. 결국 저타는 달리는 기차에서 뛰어내려 자살을 하고 마는데...
