Manhattanites follows the lives of a group of people in New York City who are faced with life changing circumstances and decisions over a period of several months. The choices they makes have far reaching consequences on not only them but people they do not even know.

Mikey and Rose are professional assassins. They are also lovers, and once upon a time Rose also knew Mikey's father. During an elaborate hit job for a malicious gangster, Rose fails to kill the main target, the gangster's pregnant girlfriend. Instead, she delivers her baby and convinces Mikey to help her protect them both. As Rose weakens from cancer, Mikey becomes breadwinner for this unusual family. But tranquility will not last.

A street artist (Charles Lane) rescues a baby girl (Nicole Alysia) after her father is murdered. The artist then sets off to find the mother, but has to first learn how to care for the child. Ultimately he ends up in a horse drawn chase of the murderers.
