Bob Harris (Bill Murray) é uma estrela de cinema, que está em Tóquio para fazer um comercial de uísque. Charlotte (Scarlett Johansson), por sua vez, está na cidade acompanhando seu marido, um fotógrafo workaholic (Giovanni Ribisi) que a deixa sozinha o tempo todo. Sofrendo com o horário, Bob e Charlotte não conseguem dormir. Eles se encontram, por acaso, no bar de um hotel de luxo, e em pouco tempo tornam-se grandes amigos.
