面接のためデトロイトに来た若い女性が深夜に予約した宿泊先に到着すると、手違いなのか既に見知らぬ男が泊まっていた。不本意ながらもそこで夜を過ごすことにするが、予期せぬ客よりも恐ろしいものが待ち受けていた。
目覚めたら出口の無い館に閉じ込められていた8人の男女。猟奇殺人犯ジグソウの新しいゲームが始まる。ゲーム開始と共に遅効性の毒ガスが館内をめぐっていく。2時間以内にゲームに勝ち抜き、解毒剤入り注射器を手に入れないと死んでしまう…。一方、刑事エリックはジグソウを捕まえる。しかしジグソウは動揺することなく、エリックと2人で話をすることを要求する。なんと、エリックの息子はジグソウに監禁されていた。果たして犯人の真の狙いとは？
