目覚めたら出口の無い館に閉じ込められていた8人の男女。猟奇殺人犯ジグソウの新しいゲームが始まる。ゲーム開始と共に遅効性の毒ガスが館内をめぐっていく。2時間以内にゲームに勝ち抜き、解毒剤入り注射器を手に入れないと死んでしまう…。一方、刑事エリックはジグソウを捕まえる。しかしジグソウは動揺することなく、エリックと2人で話をすることを要求する。なんと、エリックの息子はジグソウに監禁されていた。果たして犯人の真の狙いとは？

マサチューセッツ工科大学の学生ジェイムズ・クレイトン。彼はその優秀な成績から、卒業後の進路もPCメーカーから特別に誘いを受けるなどエリート街道を約束されていた。そんなジェイムズはある日、アルバイト先のバーでウォルター・バークという男に出会う。彼はCIAのベテラン教官でリクルート担当者。バークはジェイムズに関する情報を全て調べ上げたうえ、その能力を見込んで採用するために訪れてきたのだった。ジェイムズは悩んだ挙げ句、就職先をCIAに絞り、採用試験をクリア、晴れて訓練生となるのだが…。
