고등학교 시절을 전혀 다르게 보낸 운동만 할줄 아는 젠코(채닝 테이텀)와 공부만 할 줄 아는 슈미트(조나 힐). 하지만, 둘다 '프롬(졸업 무도회)'을 못가긴 마찬가지였다. 졸업후 경찰학교에서 다시 만난 이둘은 서로의 약점을 채워주며 둘도 없는 단짝이 된다. 의욕에 불타는 초보 경찰이었지만 결국 곱상한 외모(?)때문에 고등학교 잠복근무로 부서 이동되어 버린다. 신종 약물이 활개치는 '21 점프 스트리트'의 한 고등학교에서 그 시절과는 정 반대의 고교생활이 펼쳐진다.

어느 날 범죄조직이 관리하던 도박판이 정체불명의 도둑들에게 털린다. 세탁소 사장이 도박장의 불법적인 돈을 노리고 종업원과 그의 친구에게 강도짓을 주문한 것. 사장은 도박판의 돈을 빼돌리고 사기를 친 전적이 있는 중간 관리자 마키(레이 리오타)가 범인으로 의심받을 거라며 안심한다. 하지만 돈을 잃은 도박꾼들이 사건을 해결하기 위해 프로 킬러를 고용하면서 분위기는 일변한다. 오직 돈과 자신밖에 믿지 않는 잔혹한 킬러 잭키 코건(브래드 피트)은 일과 관련된 모든 이들을 무심하고 깔끔하게 죽여나간다.

낮에는 스턴트맨과 자동차 정비소 직원, 밤에는 범죄집단의 도주를 돕는 운전을 하는 드라이버(라이언 고슬링). 이름조차 불리지 않는 무미건조한 그의 삶에 어느 날 아이린(캐리 멀리건)이 찾아왔다. 옆집에 사는 아이린과 교감하며 따스함을 느끼던 것도 잠시, 감옥에 가 있던 아이린의 남편이 돌아오며 모든 것은 원점으로 돌아간다. 하지만 감옥에서 진 빚 때문에 협박을 받자 그녀의 행복을 지키기 위해 갱단의 요구대로 전당포 터는 일에 협조하지만 함정에 빠지고, 위험에 노출된 아이린을 지키기 위한 드라이버의 폭주가 시작된다. 숨막히는 폭력과 치열한 사투를 벌이던 그는 서서히 자신의 숨겨져 있던 냉혹한 본성과 마주하게 되는데...
