캘리포니아주 로스앤젤레스의 선셋 대로에 위치한 대저택에서 살인사건이 발생한다. 유명 여배우의 풀장에서 한 시나리오 작가가 총에 맞은 채 죽어서 발견된 것이다. 경찰이 출동한 가운데 세상 사람들은 이 사건에 눈과 귀를 기울인다. 사건이 일어나기 정확히 6개월 전. 주로 B급 영화의 시나리오를 썼던 조셉 길리스는 벌이가 좋지 않아 차까지 압류당할 위기에 처한다. 해결사들의 추격을 받던 조셉은 선셋 대로에 위치한 대저택에 숨어들었다가 그 곳의 주인이자 과거 무성영화 시절 스타인 노마 데스몬드을 만나게 된다. 노마는 조셉에게 자신이 쓴 시나리오를 보여주며 유혹하는데...
헐리웃 스타의 꿈을 안고 LA에 온 베티는 멀홀랜드 드라이브에서 일어난 자동차 사고로 이름조차 기억하지 못하는 리타를 만나 그녀가 기억을 되찾을 수 있도록 도와준다. 베티의 적극적인 도움으로 단서를 찾아가던 리타는 한 카페의 여종업원 명찰에서 다이안이라는 이름을 보고 어떤 것을 떠올리게 된다. 베티는 이 이름이 분명 리타와 관계된 사람의 이름일 것이라 말하며 다이안이라는 인물을 찾기 시작한다. 마침내 다이안의 집을 찾아간 두 여인. 그러나 그들은 곧 상상을 초월하는 끔찍한 비밀을 발견하게 되는데...
작은 시골 마을인 바인야드에서 살고있는 천하의 순진남 밥과 소문난 작업남 월트. 성격도, 외모도 180도 다른 이들 형제는 바로 허리 22cm 가량이 붙어 있는 샴쌍둥이이다. 선천적인 핸디캡을 천부적인 재능으로 승화시킨 이들은 엄청나게 빠른 속도로 햄버거를 만드는 “번개버거”의 주인이자, 스포츠에 뛰어난 재능을 보이는 이 동네 최고의 명물이다. 하지만 어느날, 공유하고 있는 간의 90%가 밥에게 있기 때문에 더 빨리 늙을 수 밖에 없는 월트는 배우가 되고자 하는 열망을 더 이상 미룰 수 없게 되고, 더 늦기 전에 헐리우드로 진출하겠다고 폭탄 선언을 한다. 밥은 월트의 일방적인 결정에 반발하지만, 3개월만 도전해보자는 월트의 설득에 마지못해 따라 나서게 되는데… 헐리우드에 도착한 밥과 월트는 모텔에서 알게 된 에이프릴의 조언을 듣고도전을 감행하지만 문전박대만 당하고, 겨우 따낸 배역도 알고 보니 포르노 영화였다. 이 사실을 알고 기겁해서 촬영장을 나오다가 그들은 인기 스타 셰어에게 발탁이 되는데… 하지만 셰어의 속셈은 출연하고 싶지 않은 TV 시리즈에 말도 안 되는 인물을 캐스팅 함으로써 방송국으로 하여금 스스로 계약을 파기하도록 하려던 것! 하지만 셰어의 의도와는 다르게 이들 형제의 인기는 하늘 높은 줄 모르고 치솟고, 드라마는 최고 시청률을 기록한다. 그러던 중, 3년 동안 인터넷으로 사랑을 키워오던 밥의 여자친구가 두 사람이 샴쌍둥이라는 것을 알게 되고 밥은 큰 실연의 상처를 받게 된다. 이에 월트는 반드시 분리 수술이 필요하다고 생각하고 자신의 생존 가능성이 희박하다는 위험을 감수하고 수술대에 오르는데…
