待望の赤ん坊誕生を5日後に控えるピーター（ロバート・ダウニー・Jr）は、身重の妻のもとに向かうため、アトランタから自宅のあるロサンゼルスへと向かう飛行機に乗ろうとしていた。しかし、たまたま出会った役者の卵、イーサン（ザック・ガリフィナーキス）のせいで、二人でヒッチハイクをすることになる。

スポーツ記者のラウル・デュークと、サモア人で弁護士のドクター・ゴンゾー。彼らはバイクレースの取材のため、トランク一杯に「治療薬」と称したあらゆるドラッグを詰め込み、一路ラスベガスへ。超一流ホテルのスウィートルームに到着した彼らは、取材そっちのけで、早速ドラッグ三昧。ホテルを荒らしまくってやりたい放題する。
