프랭크 레노는 모범수로서 6개월만 있으면 오랜 교도소 생활을 마치고 사회로 나가게 되어있다. 그는 과거 교도소내의 생사여탈의 권한을 완전히 쥐고 있는 드럼굴 소장에게서 탈옥하여 그의 비행을 사회에 알림으로써 그를 벽지 교도소로 좌천시켰었다. 남은 형기를 모범수로 채우던 레오네가 출소를 6개월 앞두고 뜻밖에 제3의 교도소로 전출된다. 그 동안 벽지 교도소에서 복수의 칼을 갈던 드럼굴이 새로 소장으로 부임한 교도소였다. 온갖 가학으로 레오네를 괴롭히지만 꿋꿋이 견딘다. 그러나, 소장을 사주를 받은 한 수용수가 면회온 애인을 강간하겠다는 협박에 탈옥을 기도하는데, 모든 것은 소장의 계획이었다.

미국립교도소는 인간으로서는 도저히 살 수 없고 동물의 본능으로 살아가야 하는 잔혹한 곳이다. 인간의 존엄성은 사라지고 오직 생존의 의미만 부여할 수 있는 곳. 론은 이곳 감옥 생활에 차츰 적응해가지만 자신이 살기 위해 남을 죽여야만 하는 삶과 죽음의 갈림길에서 갈등하게 되는데...

웨이크 필드 교도소에 새로 부임하는 헨리 브루베이커는 교도소의 실정을 알기위해 스스로 죄수처럼 가장하고 잠입한다. 죄수로 생활하면서 어느 정도 사정을 알게 된 브루베이커는 교도소 개선을 위해 힘껏 일하지만 교도소 밖 사람들의 따가운 눈총을 받게 되고 전통적으로 교도소 죄수들을 노동력으로 이용하던 사람들에게 협조도 하지 않자 그들은 모두 브루베이커를 싫어한다. 하지만 꿋꿋하게 자기 일에 최선을 다해 가던 중, 웨이크 필드 교도소에서 거의 반평생을 보낸 아브라함이란 죄수에게서 사체 유기 사실을 듣게 된다. 아브라함은 그 사실을 누설한 것 때문에 부패한 모범수들중 대장격인 휴이에게 죽음을 당하고 이에 분개한 브루베이커는 사체 발굴에 들어간다. 이에 주지사나 상원의원은 문제가 야기될까 봐 두려워하며 브루베이커에게 일을 중단하도록 압력을 넣는다. 하지만 브루베이커는 드디어 사체발굴에 성공하고 언론에 알려서 세인의 관심을 집중시키게 된다. 그 일로 브루베이커는 해임을 당하게 되고 신임 소장이 다시 부임하게 되지만 웨이크필드의 죄수들 마음엔 브루베이커가 정의의 횃불로 남게 된다.

미국 알래스카, 스톤헤이븐 중범 형무소(Stonehaven Maximum Security Prison). 탈옥수 매니와 버크는 천신만고 끝에 달리는 기차안에 올라타게 된다. 그러나 불행히도 그들이 탄 기차의 기관사가 갑자기 심장마비로 죽는다. 기차는 고삐 플린 망아지처럼 폭주하고 사태의 심각성을 알게된 철도회사에서는 다른 기차와의 충돌의 피하기 위해 노심초사하나 달리 해결할 방법이 없어 애만 태운다. 뒤늦게 기차안에 사람이 타고있는 사실을 알게 된 철도회사는 한가닥 희망을 걸고 기차를 정지시키려고 한다. 한편 탈옥수를 잡으로 온교도소장이 헬기에서 사다리를 타고 기차로 내려온다.

1960년, 샌프란시스코. 중죄수들만 수감하는 샌프란시스코만 바로 앞의 작은섬 알카트래즈 연방 교도소. 냉철하고 이지적인 죄수 모리스는 입소 첫날부터 탈옥할 생각을 하지만 악마같이 집요한 소장의 표현대로 아직 한번도 탈옥에 성공한 예가 없는 교도소답게 허점이 보이지 않는다. 그러나 모리스는 감방 건물이 낡고 바다의 염분 습기 때문에 시멘트와 철근이 많이 삭았다는 점에 착안하고 환기통을 뚫고 천정을 통해서 탈출할 계획을 세운다. 존과 클리렌스 형제와 함께 탈옥을 약속하고 환기통 구멍을 파면서 교도관들의 눈속임을 위한 위장물과 탈옥장비들을 동료 죄수들의 도움으로 하나씩 준비한다. 몇차례 위기를 넘긴 일행은 치밀한 계획과 대담한 행동으로 환기통을 통하여 교도소 건물을 빠져나간다. 다음날 아침 교도소는 비상령이 내리고 근처 바다에서 대대적인 수색 작업을 폈으나, 끝내 찾지못하고 곧이어 알카트래즈 감옥이 폐쇄되는데...
